Psilocybin is the most researched of the classical psychedelics. Mexico is its ancestral home — the Mazatec mountains of Oaxaca were the first place modern science encountered psilocybin mushrooms, through the work of María Sabina in the 1950s. Today, retreats in Mexico range from indigenous-lineage ceremonies to clinically-supervised programmes with on-site physicians.
Psilocybin is a tryptamine alkaloid found in over 200 species of mushroom, most commonly Psilocybe cubensis. In the body, psilocybin converts to psilocin, which acts primarily on serotonin 5-HT2A receptors. Effects last 4–6 hours after oral dosing.
The clinical evidence is the strongest of any psychedelic outside of MDMA-assisted therapy. Multiple randomised trials show large and durable antidepressant effects, including in treatment-resistant depression. Johns Hopkins follow-up data shows 75% of patients with major depressive disorder maintained clinically significant improvement at 12 months after two sessions.
The FDA has granted psilocybin Breakthrough Therapy designation twice — once for treatment-resistant depression and once for major depressive disorder. Phase III trials are ongoing.
Psilocybin-containing mushrooms are technically restricted under Mexican federal law, but indigenous-traditional and religious-ceremonial use is tolerated, particularly within Mazatec, Mixtec, and Huichol communities. This is a grey zone — most retreats operate in a wellness/ceremonial framework rather than a medical one.
